    Recombinant human Fibroblast Growth Factor-basic (bFGF) (AA134-288), also called as FGF-2 or HBGF-2, is a bioactive protein intended for use in cell culture applications. bFGF is a heparin-binding member of the FGF superfamily of molecules. It is involved in a number of biological processes including embryonic development, differentiation, survival, regeneration and migration (1-5). In addition, bFGF is a critical factor for growing embryonic stem cells in culture to remain in an undifferentiated state. Recombinant human FGF-basic is a 17.3 kDa protein consisting of 154 amino acid residues.

    Figure 2a.  NIH/3T3 (mouse fibroblast cell line, ATCC: CCL-1668) were cultured before adding bFGF.
    Figure 2b.  NIH/3T3 (mouse fibroblast cell line, ATCC: CCL-1668) were cultured with 0.25 ng/ml in serum-free medium at 37 °C, 5% CO2 for 24 hours.
    Figure 3.    bFGF-2 Activity Assay. The optical density (OD) of each concentration of the testing samples, which is directly proportional to the cell number, was read at 492 nm at the endpoint of incubation.     Endotoxin:
    Less than 0.01 ng/μg cytokine as determined by the LAL assay
    Bioactivity:
    ED50= 0.1-1.0 ng/ml as determined by the dose dependent proliferation of NIH 3T3 cells (figure-2, 3).
    Formulation:
    Sterile filtered through a 0.22 micron filter and lyophilized with 0.5 % BSA and Tris buffer (10 mM Tris pH7.5, 150 mM NaCl).
    Purification:
    Sequential chromatography
    Purity:
    Greater than 98% as determined by SDS-PAGE (figure-1) before adding BSA.
    Reconstitution:
    Centrifuge vial before opening. It is recommended to reconstitute bFGF in sterile 10 mM Tris-HCl, pH 7.5 or PBS to yield a stock solution of 0.1 mg/ml of bFGF. It is stable for up to 6 months when stored at -20°C and up to 12 months when stored at -80°C. Multiple freeze/thaw cycles will result in significant loss of activity.

    Storage & Stability:
    The product is shipped at ambient temperature with ice bags. Upon receipt, it should be stored immediately at -20 °C to -80 °C. Lyophilized bFGF is stable at -20 °C to -80 °C for up to 16 months.

    Useful for cell culture and for the study of signaling pathways.
